The mitogen-activated protein (MAP) kinase family is comprised of highly homologous proteins that transduce signals from tyrosine kinases to the nucleus. The serine-threonine protein kinase, MAP kinase-activated protein kinase-2 (MAPKAP-2), plays an important role in signaling by this family of proteins and was identified by its ability to mediate tyrosine phosphorylation of the R-subunits of cyclic AMP-dependent protein kinase (PKA) (see, for example, Nakahira et al., Proc. Natl. Acad. Sci. U.S.A. 90: 9747-9751 (1993)). MAPKAP-2 was subsequently shown to have more than one protein kinase-mediated target, including the insulin receptor substrate-1 (IRS-1) (see, for example, Song et al., Nature 355: 342-346 (1992); Song et al., EMBO J. 13: 709-717 (1994); and Song et al., Nature 369: 755-760 (1994)). The binding of insulin to the insulin receptor results in tyrosine phosphorylation of several intracellular substrates, including insulin receptor substrate-1 (IRS-1) and -2 (IRS-2), leading to the formation of a signal transduction complex that activates the phosphatidylinositol 3-kinase (PI3K) signal transduction pathway and is essential for insulin-induced cell growth. To date, there are no known mammalian genes that have been shown to encode MAPKAP-2. Accordingly, the use of DNA encoding MAPKAP-2 to transduce signals, for example, in methods of signal transduction (e.g., methods of treating disease), identification of novel receptors (e.g., the identification of novel insulin receptors), or for the generation of transgenic animals is not possible.Adsorption and separation of polystyrene using sil be359ba680
